There are as I have seen four cities around Biwako, like Otsu for example. I probably wont have time to visit them all so I would like some recommendations which city or place around the lake would be "best" to visit. Vill be staying in Osaka
Hikone (??) is a small city at the shores of Lake Biwa, Japan's largest lake. The city is most famous for its castle, one of only four castles in Japan designated as national treasures. Hikone makes an easy and pleasant 3-6 hour stop-over on a trip between Tokyo and Kyoto or a side trip from Kyoto.

Consider Omihachiman (45 minutes from Kyoto on the JR Biwako Line). Visit the preserved merchant houses of Omi (a 15-minute walk from the station twoards Mount Hachiman)
