It's definitely possible if you use a 3rd party tool like autohotkey, but I think it cannot be done in the game options or dota2 config. Really annoying.
25 Mar 2016 Hi, guys, after today's update I can't add unit to control group without changing composition of group. Usually to create control group I used "ctrl+hotkey group", and for further replenishment control group I used "SHIFT+hotkey group". This is very important when you play at Chen or Enchantress. Please fix it.

11 Oct 2014 To set a control group, you highlight and select (with cursor) which heroes you wish to bind. Then, when they're selected, you press and hold 'Ctrl' and press the key you wish to bind them to (such as '1' or '2' in the example above).
