24 May 2016 Reticulocyte count is performed by manual method. A few drops of blood (collected in EDTA) are incubated with new methylene blue solution which stains granules of RNA in red cells. A thin smear is prepared on a glass slide from the mixture and reticulocytes are counted under the microscope.
Counting reticulocytes. 1. Look into the slide area using 10x objective. 2. Add a drop of oil. 3. Move to 100x. 4. Count all red cells (reticulocytes + RBC) using a manual counter. 5. Make sure not to count WBC (large nucleated cell). 6. The number of reticulocytes is counted also on outside paper to be able to calculate.
Perform the Miller disc procedure using another student's set of retic slides. Find a suitable area of the smear where the cells are close together but not touching. A minimum of 112 cells should be counted in the small square because this is equivalent to 1008 red cells in the large square.
